Samples' Criteria Sample
Amount
- It is recommended that 25 to 200 pmole of protein be submitted.
- More sample provided greater the number and accuracy of amino
acids identified. In general, more sample is required when submitted
on PVDF than in solution.
Sample Form
- Sample can be prepared as solution, dried sample or electroblotted
protein on PVDF membrane.
- For solution samples, solvent used in dissolving samples should
contain only volatile components such as water, acetonitrile etc.
Please concentrate your sample in a small volume of solution.
Usually, no more than 60 Łgl of the sample will be loaded for sequencing.
- For proteins adsorbed onto PVDF, the piece of PVDF should be
no larger than 3 x 7 mm and the number of pieces no more than
five. Keep in mind that transfer is not 100% efficient so load
an adequate amount of protein or peptide onto the gel prior to
electrophoresis.
- Please note that nitrocellulose cannot be used in our sequencer.
- If your sample is supplied dried, we will try to dissolve it
into distilled water. Insoluble samples will not be sequenced.
Purity
- Sample should contain one protein component only.
- Sample should be as homogeneous as possible and free of salts,
detergents, lipids and other non-volatile ingredients (SDS, Tris
etc.). Please state the possible contaminants in the submission
form. ˇ@
