2. Physical properties

You can predict various physical properties of a protein based on its primary sequence.
 

1. Go to the ProtParam home page.

 

2. Copy the query sequence and paste it into the window, then press the 'Compute parameters' button.

3. You will see the results of the parameters calculation:

The result report has a lot of useful parameters predicted for the query sequence (e.g. pI, MW, Amino acid composition)

Q1.Its isoelectric point (pI) of the protein is predicted to be 9.80. Is it a reasonable value when you take the biological function of this protein into account?
 

Q2. The predicted extinction coefficients can be used to estimate the protein concentration by measuring the OD280. What is the protein concentration of yeast L30 sample if the measured OD280= 0.1, using a cuvette of 1 cm light path?
